摘要
目的:探讨小鼠表皮角质形成细胞(KC)中,Ca2+对组织型纤溶酶原激活剂(tissueplasminogenactivator,tPA)表达的调节及其作用机制。方法利用免疫细胞化学及RT-PCR等方法,定性、定量检测在不同浓度Ca2及异博定、放线菌素D作用下,tPA蛋白质或mRNA表达的变化情况。结果:与低Ca2+浓度相比,高Ca2+浓度下,表皮KC中tPA蛋白及mRNA的量均发生了非常显著的增加,而异博定及放线菌素D的加入,则可抑制高Ca2+浓度的这一作用。结论:在小鼠表皮KC中,胞外高浓度Ca2可通过细胞膜上的电位依赖性离子通道进入胞内,在转录水平上调节tPA的表达。
AIM:Tissue plasminogen activator(tPA) is an important proteinase in the epidermal keratinocytes.The expression of tPA could significantly increase in the wounded epidermis or in the lesioned epidermis of some skin diseases patients.In the epidermal keratinocytes, the calcium is an common regulator of the protein expression, but whether could it regulate the abnormal expression of tPA? How about the regulating mechanism? To these problem, we made our investigation.METHODS:The keratinocytes from back skin of the embryo of Kunming mouse were cultured, and then the immunocytochemistry and reverse transcriptase polymerase chain reaction were used to detect the expression of tissue plasminogen activator protein and mRNA in keratinocytes treated with calcium in different concentrations, verapamil and actinomycin D.RESULTS:The detection of immunocytochemistry and reverse transcriptase polymerase chain reaction all displayed that, comparing with low concentration calcium, under the high concentration calcium, the expression of tissue plasminogen activator protein and mRNA in keratinocytes were significantly increased, which could be inhibited by both of the verapamil and actinomycin D.CONCLUSION:In the mouse keratinocytes, the high concentration of extracellular calcium could regulate the expression of tissue plasminogen activator on the level of the transcription after its entry to the cells by potential dependent calcium ion channel. The increased concentration of calcium may be an important factor which could induce the abnormal expression of tPA in lesioned epidermis.
