摘要
目的观察脑脊液对体外培养的骨髓基质细胞的影响并探讨颅内移植骨髓基质细胞的可能性。方法分离BALB/C小鼠的骨髓细胞进行体外培养,然后将贴壁的骨髓细胞传代,在含不同比例脑脊液的培养基中培养,观察其生长情况。结果在含不同比例脑脊液的培养基中生长的骨髓基质细胞形态相似,呈多角形、梭形以及圆形;各组骨髓基质细胞计数差异无显著性意义(P>0.05)。结论骨髓基质细胞在50%(体积百分浓度)的脑脊液培养基中仍可继续生长、增殖,提示这种细胞对生长环境有高度的适应性。
Objective To study the effect of cerebrospinal fluid(CSF)on bone marrow stroma cell(BMSC)culture in vitro and the possibility of intracranial implantation of BMSC.Methods Isolated bone marrow cell from femora of BALB/C mice were cultured in DMEM:F12media,the adherent cells were har-vested,then cultured in modified media containing different ratio CSF mixture,and the status of cell prolifer-ation were recorded.Results The morphological features of BMSC cultured in media with different ratio CSF were similar with round,fusiform or astroid appearances.There were no significant differences(P>0.05)be-tween the counts of BMSC cultured in different media.Con clu sion Bone marrow stroma cell can still prolif-erate in media even containing of50%cerebrospinal fluid,thus it is indicated that these cells possess of high adaptability to their growth environment.[
