摘要 目的 :探讨不同周期运动对大鼠心肌细胞热休克蛋白 72mRNA表达的影响。方法 :以不同周期跑台运动为运动模式 ,观察了一周运动、二周运动和三周运动后 2 4h大鼠心肌细胞热休克蛋白 72mRNA的表达。结果 :安静对照组大鼠心肌细胞存在HSP72mRNA的基础结构性表达 ;一周运动后大鼠心肌细胞HSP72mRNA表达明显增多 ,两周运动后HSP72mRNA表达较一周运动稍减少 ,但无统计学意义 ;三周运动后大鼠心肌细胞HSP72mRNA表达较运动一、二周减少 ,且存在显著性差异。结论 :运动可以造成心肌细胞HSP72mRNA的表达增加 ,长时间规律运动可以使HSP72mRNA表达在细胞中累积 。 Objective To study whether exercises of different durations can effectively induce the expression of myocardial cells HSP72 mRNA. Methods SD rats were randomly divided into four groups: (1) control group, (2) training for 1 week period group, (3) training for 2 weeks period group, and (4) training for 3 weeks period group. The rats were trained on treadmill in the intensity of 75%VO 2max for 1 week, 2 weeks, 3 weeks separately. After training all rats were decapitated at intervals of 24 hr and HSP 72 mRNA expression of cardiac myocytes was detected by reverse transcription-polymerase chain reaction (RT-PCR). Results (1) HSP72 mRNA can be basically expressed in rats of control group. (2) There is a significant increase of HSP72 mRNA in myocardial cells in 1-week-period group compared with control. (3) A decrease of HSP72 mRNA was observed in 2-week-period group compared with 1-week-period group,but there was no statistical difference between the two groups. (4) The decrease of HSP72 mRNA was more pronounced in 3-week-period group than in 2-week-period or 1-week-period group. Conclusion Exercise is a sufficient stimulus to induce or enhance the synthesis of heat shock and/or stress proteins in myocardial cells. HSP72 mRNA can accumulate in cells after long time exercise, but the accumulation decreases gradually after some time.
