摘要
目的 :观察柴胡对肝星形细胞有关胶原降解基因表达的影响。方法 :体外培养肝星状细胞 ,采用MTT法选择药物的实验浓度 ,逆转录多聚酶链反应检测间质性胶原酶及金属蛋白酶组织抑制因子 -1基因表达水平。结果 :与对照组比较 ,柴胡组金属蛋白酶组织抑制因子 -1基因表达水平明显降低(P<0.01) ,而间质性胶原酶基因表达水平无明显差异(P>0.05)。结论 :柴胡的抗肝纤维化作用可能部分是通过抑制金属蛋白酶组织抑制因子
Objective:To study the effects of bupleurum on collagen degradation of hepatic stellate cell in vitro.Methods:Hepatic stellate cell line(HSC_T6)was cultured in vitro.Concentration of bupleurum was selected by MTT colorimetric assay.The gene expression levels for interstitial collagenase and tissue inhibitor of metalˉloproteinase_1(TIMP_1)were measured by reverse transcription polymerase chain reaction.Results:Compared with control group,the gene expression level of TIMP_1in bupleurum group significantly decreased(P<0.01),and there was no significant difference between two groups in gene expression level of interstitial collagenase(P>0.05).Conclusion:Bupleurum could prevent liver fibrosis by inhibiting gene expression of TIMP_1.
