常见性病患者血液套式PCR扩增HCMV-DNA的研究 被引量:1

摘要 目的 建立检测人巨细胞病毒 (HCMV)感染的套式 PCR方法并探讨常见性病患者 HCMV的感染情况。方法 在 HCMV IEA基因内自行设计两对引物 ,建立套式 PCR检测 688例常见性病患者及 1 38名健康献血员血液HCMV- DNA,同时应用捕获 ELISA检测 HCMV- Ig M。结果 性病患者 HCMV- Ig M及 DNA阳性率分别为 34.74%和39.53% ,与对照组 (8.0 0 %、1 0 .1 4 % )相比均有显著性差异 (P<0 .0 1 )。从性病类型来看 ,梅毒患者 HCMV感染率最高 ,其 HCMV- Ig M及 DNA阳性率分别达 46.80 %、51 .90 % ;从年龄分组看 2 6~ 35岁组为 HCMV感染高峰组 ,HCMV- Ig M及 DNA阳性率分别达 40 .2 0 %、46.62 %。结论 性病患者易发生活动性的 HCMV感染 ,性接触也是成年人之间 HCMV的重要传播途径之一。套式 PCR是一种临床检测 HCMV的快速有效手段 ,值得临床推广。 Objective To establish the nested PCR method for detection of HCMV infection in patients with sexually transmitted diseases.Method HCMV DNA and anti HCMV IgM were simultaneously tested by nested PCR using two primer sequences from HCMV IEA genome and capturing ELISA respectively in sera from 688 patients with sexually transmitted diseases and 138 blood donors.Results Positive rates of HCMV IgM and HCMV DNA in 688 patients with sexually transmitted diseases were 34.74% and 39.53% respectively,there was a significant difference when compared with control groups(HCMV IgM 8.00% and HCMV DNA 10.14%)(P<0.01).In terms of classification of sexually transmitted diseases,patients with syphilis had a peak incidence in HCMV infection,in which the positive rate of HCMV IgM and HCMV DNA were 46.80% and 51.90% respectively.In terms of age,the group of 26~35 years was peak group of HCMV infection,in which HCMV IgM positive rate was 40.20% and HCMV DNA 46.62%.Conclusion Active HCMV infection easily occurred in the patients with sexually transmitted diseases,and sexual behavior also was one of the important transmitted way among adults.Nested PCR would be useful of rapid detection of HCMV in clinic.
