胆囊癌组织中抑癌基因p53突变与肿瘤血管生长的相关性研究

摘要 目的:探讨胆囊癌中抑癌基因p53突变与肿瘤血管生长的相关性。方法:1998年1月至2000年12月共收集44例胆囊切除标本,其中24例胆囊癌,20例胆囊腺瘤。用免疫组化方法定性检测突变抑癌基因p53蛋白产物的表达,同时检测标本中血管内皮生长因子(VEGF)的表达以反映肿瘤血管生长的情况,比较二者的相关性。结果:胆囊癌中p53蛋白的阳性率达到62.5%。VEGF的阳性率为75%,其阳性表达率随胆囊癌的病理分期的进展而逐渐增强(P<0.05)。p53与VEGF的阳性率显著相关(P<0.01)。结论:抑癌基因p53突变在胆囊癌早期发生中起重要作用,在胆囊癌晚期则通过促进肿瘤血管的生长,导致癌肿的浸润转移。 Objective:To explore the relationship between gene p53mutation and carcinoangiogenesis in gallbladder cancer.Methods:The expression of the tumor suppressor gene p53was measured using immunohistochemical assay in24gall-bladder cancer and20gallbladder adenoma specimens.Expression of the vascular endothelial growth factor(VEGF)was measured in these specimens to reflect the carcinoangiogenesis.The relationship between them was then evaluated.Re-sults:The positive expression rate of p53was62.5%in cases of gallbladder cancer,while that of VEGF was70.8%.The positive expression rate of VEGF increased along with the up-staging of tumor(P<0.05).Positive correlation was found be-tween the expression of p53and VEGF(p<0.01).Conclusions:p53mutation facilitates carcinogenesis,resulting in gall-bladder cancer metastasis and migration.
