摘要
探讨了应用PCR和RT PCR技术对 4种主要的对虾病毒进行检测的方法 ,同时使用该方法检测了对虾天然饵料———卤虫中的 4种对虾病毒。结果显示 ,含病毒核酸的阳性对照样品分别扩增出了大小为 82 4bp、70 5bp、2 6 0bp和 2 16bp的预期产物 ,但未能从卤虫样品中检测到此 4种病毒的存在。本文报道的病毒检测方法具有快速、灵敏、准确的特点 ,可以用于进出口贸易中对活体或冰冻对虾、虾苗、对虾饵料等进行相关对虾病毒的检疫 。
White spot syndrome virus (WSSV), Infectious hypodermal and hematopoietic necrosis virus (IHHNV), Yellow head virus (YHV) and Taura syndrome virus (TSV) of shrimp are four major viruses which can cause mass mortalities of cultured shrimps. It is an important method to detect these viruses in artermia, the natural food of shrimps,and to prevent the spread of virus diseases. In this study, the technologies of PCR and RT PCR were developed and applied to detect these viruses in nine samples of artermia. Positive control samples which contained DNA or RNA of WSSV, IHHNV, YHV and TSV yielded evident amplification products showing the expected DNA fragments of 824 bp, 705 bp, 260 bp and 216 bp respectively. But none of these four kinds of viruses can be found from samples of artermia and negative controls. The methods reported here can be used to quarantine WSSV, IHHNV, YHV and TSV in live or frozen shrimps, larva and feed of shrimp in the fields of import and export.
