趋化因子及其受体在HIV感染中的作用 被引量:1

摘要 随着趋化因子及其受体在HIV感染中的作用被发现,人们对HIV如何进入细胞有了进一步的认识。CD_4分子是HIV进入靶细胞的主要受体,但仅有CD_4尚不足以使HIV侵入细胞,还必须以趋化因子受体为辅助受体。辅助受体与CD_4分子形成复合体,gp120与之结合并发生构象改变,使gp41暴露,从而引发膜融合。HIV利用靶细胞的辅助受体随感染阶段而不同,初期以被嗜巨噬细胞病毒利用的CCH5为主,随着HIV由巨噬细胞嗜性转变为双嗜性和T细胞嗜性,辅助受体也从CCR5转变为以CXCR4为主。利用阻断趋化因子受体的方法可以有效地阻止HIV入侵细胞,从而为艾滋病的防治开辟了新的前景。本文就趋化因子及其受体的结构和功能,HIV感染细胞及其与辅助受体相互作用的机制,辅助受体基因多态性与艾滋病的进程,以辅助受体为靶位的新药开发作一综述。
