机械牵张对培养的心肌细胞分泌AngⅡ、ET-1及AngⅡ对培养的成纤维细胞分泌ET-1的影响

摘要 目的 探讨机械牵张对培养的心肌细胞分泌血管紧张素Ⅱ(Angiotensin Ⅱ,Ang Ⅱ)、内皮素-1(Edothelin-1,ET-1)的影响,及AngⅡ对培养的成纤维细胞分泌ET-1的影响。方法 在可牵张硅胶膜上培养心肌细胞,通过放免法观察10min的牵张刺激对心肌细胞分泌AngⅡ、ET-1的影响,并观察AngⅡ对培养的成纤维细胞分泌ET-1的影响。结果 牵张10min后,CM培养液中AngⅡ和ET-1的浓度明显升高。结论 机械牵张使CM快速分泌AngⅡ和ET-1,前者使培养的成纤维细胞分泌ET-1增加。 Objective To study the effects of stretch on Ang Ⅱ and ET - 1 secretions from cultured cardiac myocytes ( CM ) and the effects of angiotensin Ⅱ (AngⅡ) on endothelin- 1 (ET- 1) secretions from cultured myocardial fibroblasts (MF) .Methods All CMs were cultured on deformable silica gel membrane. Media were collected after exposure of CM to stretch for 10 minutes. All MF were cultured in DMEM medium with and without Ang Ⅱ . Ang Ⅱ and ET - 1 were assayed by radioimmunoassay. Results Ten minutes after stretch, the concentrations of Ang Ⅱ and ET - 1 in the medium increased remarkably. the concentration of ET - 1 rose markedly in the medium with Ang Ⅱ in which MFs were cultured. Conclusions Mechanical stretch may quickly induce the secretion of Ang Ⅱ and ET - 1 from CM and Ang Ⅱ may induce the secretion of ET - 1 from MF.
