摘要
目的方法 :通过血清药理学方法 ,选择不同浓度的含化痰散结方的兔血清 ,与人肺癌细胞SPC A1共同孵育不同时间后 ,应用倒置相差显微镜和荧光显微镜对细胞生长和凋亡状况进行形态学观察 ,应用放射免疫的方法检测SPC A1细胞内cAMP、cGMP浓度。结果 :人肺癌细胞经含化痰散结方的兔血清作用后 ,由贴壁生长而逐渐脱落 ,外形变圆 ,体积缩小 ,折光性增加 ;荧光镜下可见细胞核破碎、裂解为大小不等的凋亡小体 ;SPC A1细胞内 12小时及 2 4小时cAMP、cGMP浓度无明显变化 ,36小时、4 8小时cAMP浓度升高 ,cGMP浓度无明显变化 。
After incubating SPC Al and traditional Chinese medicine formulae Huatan Sanjie borne rabbit serum, at which different concentration we chose with serologic pharmacological method, observed and detected the growth and apoptosis of cells, and determined the concentration of intracellular cAMP and cGMP by irradiation immunity method. Results: By the action of drug borne rabbit serum, SPC A1 grew against the wall and then peeled off gradually; fluorescence microscope showed that nucleus broke into pieces, and cell split into apoptosis bodies in different size. The level of intracellular cAMP concentration, treated with drug borne rabbit serum at 36h?48h, was significantly higher than that of the control groups, but there were no significant difference in intracellular cAMP concentration between drug borne rabbit serum groups and control groups at 12h?24h. There were no significant difference in intracellular cGMP concentration between drug borne rabbit serum groups and control groups at 12h?24h?36h?48h. So ratio of cAMP/cGMP were significantly increased at 36h?48h.
