摘要
根据McClure偶联酶反应理论,通过实验对偶联酶法检验L-天冬酰胺酶活力的反应条件进行了动力学分析,确定了合理的附加酶用量,建立了偶联酶法检验此酶活力程序。用该程序测定的底物K_m值和产物的抑制常数K_1值与文献值相符。
A kinetic analysis of couple enzyme assay for L-asparaginase was made on the basis of Mc-Clure's theory. A correct amount of auxiliary enzyme (Glutamate Dehydrogenase) was calculated. A procedure of continuous assay for L-asparaginase with GDH as a couple enzyme was designed. As an application of the couple enzyme assay, the determination of apparent K_mof L-asparagine and K_1of L-aspartic acid was carried out.
