摘要
氨基酸分离常用茚三酮作显色剂,以滤纸作为载体,通过选择合适展开剂的配比和合理的显色时间,让样品溶液在滤纸上层析达到分离的目的。在试样及展开剂的性质,滤纸的性质,展开时的温度、方式和距离相同的条件下对半胱氨酸、谷氨酸、L-脯氨酸等三种混合物的纸上层析方法和分离效果进行了研究;并介绍了点样、展开方式及Rf值计算等,讨论了展开剂的配比中正丁醇、冰醋酸及水对分离效果的影响。得出了三种氨基酸的最佳分离条件,即试样为0.2%的混合物的HCl溶液;展开剂正丁醇∶冰醋酸∶水的体积比为4∶1∶1,用茚三酮的乙醇溶液为显色剂。
3dioxohydrindene is usually used as indicator and filter paper as carrier in separation of amino acids. The sample solution goes through the filter paper when the right developer's proportion and reasonable developing time are selected. The authors study the method of paper delaminating separation of cysteine, glutamic acid and Lproline and its effect under the condition of the same nature of the test sample, developer and filter paper, and same developing temperature, method and distance. The authors also introduce spot sample analysis, developing method and Rf calculation, and discuss the effect of the proportion of nbutanol, acetic acid and water in the developer to the separation. Finally, the best separate condition are received,in which the test sample is 0.2% HCl solution, the proportion of nbutanol, acetic acid and water in the developer 4:1:1,and indicator 3dioxohydrindene's ethanol solution.
