摘要
从国产玫瑰红链霉菌336菌体,经制成丙酮干粉、抽提、DEAE-52纤维索柱层析、硫酸铵分级、DEAE-Sephadex A-50柱层析、Sephadex G-200柱层析后得到电泳纯酶。对纯化酶的分子量、氨基酸组成、热稳定性等性质进行了研究。
Glucose isomerase (GI) in the cells of St. Rosechromogenus No.336 was isolated by extracting GI from acetone freeze-drying powder. The GI was purified successively by means of DEAE-cellulose-52 column chromatography,ammonium sulfate fractionation, DEAE-Sephadex A-50 column chromatography,and Sephedex G-200 column chromatography. The amino acids composition,molecular weight, thermostability, pH stability, CD spectra of the isolated enzyme were determined.
