摘要
目的 应用高效毛细管区带电泳法测定脑瘫患儿血清中 6种氨基酸含量。 方法 采用以 2 ,4 -二硝基氟苯作为衍生剂的高效毛细管区带电泳法测定血清中 6种必需游离氨基酸的含量。 结果 6种必需氨基酸的线性范围为 5~ 5 0 0 μm ol/ L,相对标准偏差为 3.6 %~ 9.5 % ,平均回收率为 91.2 %~ 10 5 .0 % ;脑瘫患儿血清 6种氨基酸的含量与正常对照组相比 ,赖氨酸、蛋氨酸、苯丙氨酸明显降低 ,色氨酸、异亮氨酸、缬氨酸无明显改变。 结论 高效毛细管区带电泳可以准确测定血清 6种必需氨基酸的含量 ;
Objective\ To study the application of high performance electrophoresis capillary method for determination of six amino acids in serum of cerebral palsy infant.\ Methods\ A rapid method for the determination of free amino acids in serum was developed.\ The assay is based on precolumn derivation of amino acids with 2,4\|dinitrofluorobenzene, separation of the derivatives by capillary zone electrophoresis.\ Results\ The linear range of six free amino acids was 5~500 μmol/L, and the relative standard deviation was 3.6%~9.5%.\ The average recovery was 91.2%~105.0% in the determination of six amino acids in serum.\ Compared with the normal group the assay of lysine, methionine and phenylalanine in the serum of cerebral palsy was reduced remarkably, while tryptophan, isoleucine and valine shows insignificant changes.\ Conclusion\ The high performance capillary electrophoresis can be used for determination the content of human body six essential amino acids accurately in the serum.\;
