摘要
目的 :观察犬膨体聚四氟乙烯人工血管补片移植 ,在不同时期的组织形态学变化。方法 :用e -PTFE补片修补 16条犬的腹主动脉 ,在植入后 2、4、8、12、2 0、2 8周采集标本 ,作光镜、扫描及透射电镜观察。结果 :①内皮细胞 2周已在材料上出现 ,8周时完全覆盖材料面 ;②内膜增生主要是平滑肌细胞和胶原纤维增生 ;③在 8周以前内膜增厚较快 ,12周以后逐趋缓和 ;④补片与血管吻合处增生明显 ;⑤增生的内膜容易与材料分离。结论 :内皮细胞可以覆盖e -PTFE材料表面 ,其内膜增生主要是平滑肌细胞和胶原纤维增生引起 ;主要原因可能是内皮细胞慢性损伤 ,血小板聚集 ,血流动力学改变 ,材料与血管顺应性不一致等。
Objective:To observe the cell coverage of the e-PTFE patch,the cellular proliferation and morphologic change of neointima. Methods: Sixteen dogs′ abdominal aorta repaired by e-PTFE patches were taken as specimens at 2,4,8,12,20,28 week after implantation ,and the specimens were observed by light microscope,scanning electron microscope and transmission electron microscope. Results: ①Endothelial cells began to appear on the surface of e-PTFE patch at the second week and covered the entire patch surface at the eighth week.②Smooth muscle cell and collagen fibers formed intimal hyperplasia.③The intimal thickening was fast before eight week,but slow after twelve week.④The cells at the anastomotic site continued to proliferate.⑤The neointima over e-PTFE was easy to separate from the material. Conclusion: These results indicate that endothelial cells can cover the surface of e-PTFE patch.The intimal thickening is caused by the proliferation of the smooth muscle cell and collagen.The proliferation might be due to chronic endothelial cell injury,platelet accumulation,hemodynamic change and graft-artery compliance mismatch.
