摘要
目的 探讨膀胱移行细胞癌基因组 (CA)n重复序列不稳定性与肿瘤生物学特性的相关性。方法 2 6例膀胱移行细胞癌肿瘤基因组DNA ,Inter SSRPCR扩增 ,聚丙烯酰胺凝胶电泳。结果 PCR扩增产物约 2 0多种 ,片段大小为 1 0 4~ 1 2 0 0bp ,带型的差异主要是增加或减少 ;染色增强或减弱 ,不稳定系数与肿瘤的分级呈正相关。结论 膀胱移行细胞癌基因组中平均有 1 2 .8%的 (CA)n重复序列DNA片段与正常组织基因组有差异 ,肿瘤分级越高差异越大。
Objective To investigate the relation between genomic (CA)n sequence repeats instability and biologic characteristics of bladder cancer.Methods 26 cases of transitional cell carcinoma of bladder were researched.Inter SSR PCR has been amplificated.Polyacrylamide gels electrophoresis was made.Results PCRs yield a set of approximately 20 products.The DNA sequences were 104~1200 bp.Gains,losses and intensity changes were showed .The instability index was significantly correlated with the tumor grade.Conclusions 12.8% of genomic (CA)n sequence repeats DNA in transitional cell carcinoma of bladder is different from normal genomic.The bigger difference it was,the higher the tumor grade was.
