白纹伊蚊和埃及伊蚊防御素(defensin)成熟肽基因克隆及结构推测 被引量:5

GENE CLONING OF MOSQUITO DEFENSIN MATURE PEPTIDE AND CONJECTURE OF THE SECONDARY AND TERTIARY STRUCTURE

摘要 应用PCR技术从白纹伊蚊和埃及伊蚊基因组中扩增出defensin成熟肽基因并对其分子生物学特性进行分析 ,发现埃及伊蚊 (BoraBora株、海口株 )、白纹伊蚊 (成都株、宜兴株、广州株 )推导的氨基酸序列与埃及伊蚊defensinA相同 ;通过Chou Fasman预测法对defensinA成熟肽的二级结构进行预测 ,可以看出在defensin成熟肽氨基酸序列的 15~ 2 3位有一个α 螺旋 ,35~ 39位有一个 β 折叠 ,32~ 34位有一个转角 ; Mature peptide gene of defensin was amplified from Aedes aegypti and Ae.albopictus genome and the molecular biological feature of them was analysized.The results indicated that the deduced amino acid sequences of Ae aegypti Bora Bora and Haikou strain, Ae.albopictus Chengdu,Yixing and Guangzhou strain amplified fragment belonged to defensin.The secondary structure of mosquito defensin A conjectured by computer shows that there are an amphipathic α helix consisting of residues 15 to 23,a β sheet comprising residues 35 to 39 and a turn involving residues 32 to 34.We also conjected the tertiary structure of defensin A by homogeneity analysis.
