人CD4^+CD25^+T细胞和CD4^+CD25^-T细胞蛋白质组的双向电泳分析 被引量:3

摘要 目的 用双向电泳技术分析人CD4+ CD2 5 + T细胞和CD4+ CD2 5 -T细胞表达的蛋白质组 ,建立二者之间的差异表达谱。方法 从人脾脏分选CD4+ CD2 5 + T细胞和CD4+ CD2 5 -T细胞 ,用双向电泳技术分析两种细胞蛋白质组的表达差异。结果 分选细胞纯度分别为CD4+ CD2 5 + T细胞 82 .3 % ,CD4+ CD2 5 -T细胞 96.5 % ,双向电泳分析发现有 4个蛋白点明显仅于CD4+ CD2 5 + T细胞检测到表达 ,有 9个蛋白点明显仅于CD4+ CD2 5 -T细胞检测到表达。结论 CD4+ CD2 5 + T细胞和CD4+ CD2 5 Objective To comparatively analyze the proteomes of human CD4 +CD25 +T cells and CD4 +CD25 -T cells. Methods CD4 +CD25 +T cells and CD4 +CD25 -T cells were isolated from human spleen and their purity was identified. The difference of protein expression in CD4 +CD25 +T cells and CD4 +CD25 -T cells was analyzed by two dimensional electrophoresis. Results The purity of CD4 +CD25 +T cells and CD4 +CD25 -T cells was 82.3% and 96.5%, respectively. By means of two dimensional electrophoresis, there were 4 protein spots exclusively detected in CD4 +CD25 +T cells while 9 protein spots in CD4 +CD25 -T cells. Conclusion There is significant difference between proteomes of human CD4 +CD25 +T cells and CD4 +CD25 -T cells.
