摘要
目的 :探讨连翘提取物抑制内毒素诱导的炎症反应的作用机理。方法 :(1)采用水提醇沉法制备连翘提取物 ,将连翘制剂倍比稀释后分别与一定量内毒素充分混合 ,采用基质显色法测内毒素浓度。(2)采用致病大肠杆菌制造大鼠腹膜炎模型 ,分别采用庆大霉素、连翘提取液及连翘 +庆大霉素进行治疗 ,观察各组动物血浆中IL_1、TNF_α水平及其肝组织TNF_α、IL_1mRNA原位杂交结果。结果 :连翘31.25g/L浓度以上时即有显著拮抗内毒素的作用。连翘组与连翘 +庆大霉素组血浆中IL_1、TNF_α水平和肝组织TNF_α、IL_1mRNA表达水平低于庆大霉素组和腹膜炎模型组 ,但该两组间无显著性差异。结论 :连翘具有明确的拮抗内毒素的作用且可显著抑制细菌内毒素诱发的炎性因子的过度表达 ,其效果明显优于单纯使用庆大霉素 。
Objective:To experimentally study the inhibitory effect of Forsythia extract on inflammatory reˉsponse induced by endotoxin.Methods:(1)The Forsythia extract was produced first,and then it was diluted to different concentrations.These diluted agents were mixed with identical endotoxin,and the concentrations of endotoxin in mixtures were detected after several minutes.(2)The rat models of peritonitis caused by Esˉcherichia coli were treated by Gentamicine,Forsythia extract and Gentamicine plus Forsythia extract respecˉtively.The levels of IL_1and TNF_αin plasma and the results of in situ hybridization of IL_1and TNF_αmRNA of liver tissue were observed.Results:When the concentration of Forsythia extract was above31.25g/L,it can inhibit endotoxin markedly.The levels of IL_1and TNF_αof plasma and liver tissue in Forsythia group and Gentamicine plus Forsythia group were lower than those in Gentamicine group and peritonitis group,but there was no difference between the Forsythia group and the Gentamicine plus Forsythia group.Conclusion:The Forsythia extract can be against endotoxin and inhibit the overexpression of cytokines induced by endotoxin evidently.Its effect of against endotoxin is superior to Gentamicine.When the Forsythia extract is combined with Gentamicine,its effect is not enhanced.
