摘要
目的 :应用大鼠腹主动脉狭窄心肌肥厚模型 ,观察血管紧张素转换酶抑制剂 (ACEI)卡托普利防治心肌肥厚的作用 ,以及该作用与心肌组织内儿茶酚胺、氧自由基、血管紧张素 (AngⅡ )含量和心肌肌球蛋白重链(MHC)基因表达的关系。方法 :取体重 15 0~ 2 2 0 gSD大鼠 2 0只 ,随机分成 3组 :对照组 (7只 )、心肌肥厚组 (6只 )、心肌肥厚加卡托普利组 (卡托普利组 ,7只 )。卡托普利组大鼠每天服用卡托普利 6 0mg/kg ,药粉末溶于饮水中。 3组动物均于手术后 8周处死。结果 :①卡托普利组大鼠全心重 (HW )、左心室重 (LVW )、HW /体重 (BW )、LVW /BW各指标与心肌肥厚组比较均明显降低 ;②卡托普利可明显抑制去甲肾上腺素、多巴胺含量的降低 ,以及肾上腺素含量的增高 ;③卡托普利可增强心肌组织超氧化物歧化酶 (SOD)和谷胱甘肽过氧化物酶 (GSH Pχ)活性 ,减少过氧化脂质 (LPO)的生成 ;④卡托普利可降低压力超负荷所致的心肌肥厚组织AngⅡ含量的增高 ;⑤卡托普利可使心肌α MHC基因表达增强 ,β MHC基因表达减弱 ,并提高α MHCmRNA/β MHCmRNA比值。 结论 :卡托普利能有效地防治心肌肥厚的发生。其作用机制不仅与卡托普利调整和改善心脏局部儿茶酚胺、氧自由基代谢有关 ,而且还与卡托普利直接抑制体内肾素
Objective:To observe the effect of captopril on of myocardial hypertrophy, and it's relationship with MHC genes expression and metabolism of cathcholamines, oxygen free radicals in myocardial tissue.Methods:Twenty SD rats were divided into 3 groups: control group (7 rats), hypertrophy group (6 rast) and captopril group (7 hypertrophied rats) treated with captopril 60 mg/kg daily. All rats were killed after 8 weeks.Results:①HW, LVW, HW/BW and LVW/BW were significantly lower in those hypertrophy group after using captopril treatment. ②Captopril effectively prevented the depletion of myocardial horepineprenine (NE)?dopamina (DA) contents and elevation of epinephenine (E) content that occurred in hypertrophy group. ③Captopril promoted myocardial SOD, GSH Px activities and decreased LPO content. ④Captopril significantly prevented the increase of myocardial angiotensinⅡ content. ⑤Captopril partly reversed the decrease in α MHC mRNA expression and the increase in the β MHC mRNA expression, and raised the radio of α MHC mRNA/β MHC mRNA.Conclusion:Captopril may effectively prevent and treat myocardial hypertrophy. The effect of captopril on antihypertrophy was not only associated with regulating and improving the metabolism of catecholamines, oxygen free radicals in heart, but also closely associated with inhibiting the metabolism of RAS, and reversing cardiac MHC isoform expression.
