摘要
目的 :研究沥水调脂胶囊抑制内皮细胞凋亡的机制 ,以氧化型低密度脂蛋白诱导人脐静脉内皮细胞凋亡 ,观察Bcl -2、Bax、P5 3和Caspase -3蛋白表达。方法 :用流式细胞术检测细胞凋亡并进行周期分析 ;用细胞免疫荧光标记结合流式细胞术检测Bcl-2、Bax、P5 3和Caspase -3蛋白表达水平。结果 :沥水调脂胶囊对氧化型低密度脂蛋白诱导的内皮细胞凋亡有抑制作用 ,在此过程中沥水调脂胶囊上调了Bcl -2蛋白表达水平 ,对Bax蛋白表达未见明显影响 ,同时下调了P5 3和Caspase -3蛋白表达水平。结论 :推测沥水调脂胶囊可能是通过上调Bcl -2蛋白表达、下调P5 3和Caspase -3蛋白表达抑制内皮细胞凋亡的。
Objective:To investigate the mechanism of li shui tiao zhi capsule(LSTZ) on apoptosis of endothelial cells induced by oxidized low density lipoprotein and expression of Bcl-2,Bax,P53 and Caspase-3.Methods:The apoptosis cells were determinde by flow cytometry.The content of Bcl-2,Bax,P53 and Caspase-3 protein were analyzed by immunofluoresence staining and flow cytometric analysis.Results:LSTZ could inhibit apoptosis of endothelial cells induced by ox-LDL,and upregulated the level of Bcl-2 protein.However expression of P53 and Caspase-3 were downregulated after treatment with ox-LDL.Conclusion:LSTZ upregulated the protein expression of Bcl-2 and downregulated P53 and Caspase-3 protein expression,which inhibit apoptosis of endothelial cells.
