摘要
经皮给药是一种新兴的给药方式。判断一种药物是否适合制成经皮给药制剂 ,首先要进行大量的体外透皮实验 ,实验的目的之一是为了获得描述药物透皮性质的参数 ,其中最重要的参数就是药物的透皮系数。在目前所见到的文献中 ,研究人员的工作重心大都放在实验工作上 ,对于实验数据的处理与分析 ,还没有形成一套固定的、合理的数学方法。本文的目的是通过对观测数据的分析 ,建立一种恰当的数学模型 ,并通过对这一模型中参数的估计来得到描述药物透皮性质的重要参数 ,非线性回归模型被用来完成这一工作。在最后的讨论中发现 ,本文的工作进一步支持了将皮肤视作单层渗透膜的理论。
Transdermal drug delivery system is a new way of drug delivery. A great amount of experiments in vitro was needed to judge whether a drug is suitable for transdermal delivery. One purpose of these experiments was to get the parameters that describe the drugs' transdermal characters. Penetration coefficient is one of the most important. In the literature, most of the work was concentrated on experiments. There isn't a reasonable method on how to deal with and analyze the experimental data. After the analysis of the experimental data, a suitable mathematical model is presented in this paper. The important parameters for the description of the drugs' transdermal character were obtained by estimating the parameters of the model. Nonlinear regression model was chosen for this purpose. Our work supports the theory that the skin could be regarded as a singular osmotic membrane.
