摘要
目的 研究细胞外调节蛋白激酶 ( ERK)在三尖杉酯碱 ( HRT)诱导 Jurkat淋巴瘤细胞凋亡中的作用。方法 用四甲基偶氮唑盐 ( MTT)法、流式细胞术和 Western印迹检测法分别检测细胞增殖抑制率、细胞凋亡和磷酸化ERK蛋白的表达。结果 研究发现下调磷酸化状态的细胞外调节蛋白激酶的表达可以增强淋巴瘤细胞系 Jurkat对三尖杉酯碱的敏感性。结论 ERK通路对维系 Jurkat淋巴瘤细胞生存有重要作用 。
Objective Study the roles of extracellular regulated protein kinases (ERK) in Jurkat lymphoma cell apoptosis induced by harringtonine (HRT). Methods The proliferation inhibition rates were measured by MTT method, cell cycle and apoptosis were analyzed by flow cytometry and phosphorylated ERK 1/2 protein expression was detected by Western blot method. Results The down-regulation of phosphorylated ERK 1 and ERK 2 protein exprssion levels could enhance the chemotherapeutic sensitivity of Jurkat lymphoma cells to HRT. Conclusion ERK signal transduction pathway may play an important role in survival of Jurkat lymphoma cells. The inhibition of ERK signal transduction pathways can enhance the sensitivity of Jurkat lymphoma cells to apoptotic inducer.
