摘要
通过多种重原子微扰剂和实验条件的选择,成功地实现了C6SCl在数种基质上的RTP发射。发现Pb(Ⅱ)盐是这种RTP发射的最有效的外部重原子微扰剂。在滤纸基质上,以1mol/L Pb(Ac)_2作重原子微扰剂获得了强度高且信/背比亦高的RTP信号,λ_(ex)/λ_(em)=320/496nm。0.2μl点样体积中,C6SCl含量在4~200pmol范围内与RTP信号强度呈良好的线性关系。
Recently, the 6-coumarinsulphonyl chloride (C6SC1) was used as fluorescence and low temperature phosphorescence labeling reagent for amino acid. In this paper, the room temperature phosphorescence (RPT) of C6SC1 on the several solid substrates, such as filter paper, films of polyamide, microcrystalline cellulose, cellulose nitrate and silica gel for chromatography etc. was induced successfully by selection of the heavy atom perturbers and experimental conditons. It is found that the RTP emission of C6SC1 has high selectivery, only two-valent lead salts are efficient perturber in the fifteen compounds and the Pb(Ac)_2 induced the most strong RTP signal. Using 1.0mol/L Pb(Ac)_2 as perturber and filter paper as solid substrate, more intensive RTP signal with high signal to blank ratio was obtained. The RTP signal intensity (λ_x/λ_m=320/496nm) as a function of the amount of C6SC1] has a good linear relationship in 0.2 μl sample solution containing 4 to 200 pmol. These results mean that C6SC1, a new developing luminescencelabeling compound, can be expected to serve as not only a fluorescence labeling reagent, but also a RTPone.
