摘要
将尿酸氯化酶和辣根过氧化物酶通过戊二醛共价交联,固定在3-氨丙基多孔硅胶上,作成光纤尿酸生物传感器的传感膜。在传感膜微环境中,尿酸的酶催化氧化反应同鲁米诺的酶催化氧化发光反应相偶合,产生化学发光信号。尿酸在0.1~5μg/ml范围内与发光信号值呈线性关系。寿命为60天。
A fiber optical bioscnscr responding to uric acid is prepared by covalcntly coupling uricase and horseradish pcroxidase to 3-aminopropyl porosity silica via glutaraldehyde and attaching the Silica to the end of fier-optic bundle. In the microenvironment of the membrane, the uricase catalyzes the oxidation of uric acid to produce hydrogen peroxide, which then reacts with luminol under the catalysis of horseradish peroxidase to produce chemiluminescence. The light intensity relates to the concentration of uric acid in the sample solution. The linear range for uric acid is 0.1~5μg/ml. This biosensor has been used to determine the uric acid in serum and urine. The results are in agreement with those obtained by current method.
