MTT法癌细胞体外药敏试验的改良及其指导下胃、肠癌的预见性化疗 被引量:3

摘要 目的 研究 MTT法体外药敏试验的改良方法及其指导下胃肠癌的预见性化疗。方法 取 2 7例新鲜胃肠癌标本经梯度离心后行体外 MTT法药物敏感试验 ,设为 MTT试验组 ;2 7例同期胃肠癌患者作经验对照组。试验组按药敏试验结果化疗 ,耐药者按临床经验化疗 ;对照组按临床经验化疗。结果  2 7例药敏试验完成实验 ,药敏阳性者胃癌 8例 ,肠癌 5例 ;阴性者胃癌 7例 ,肠癌 7例。临床化疗结果 :试验组中对药物敏感者化疗有效率76 .9% ,耐药者中有效率 14 .3% ,差异有非常显著性 ,(χ2 =10 .7,P<0 .0 1) ,试验组总有效率为 4 4 .4 % ,经验对照组化疗后有效率为 2 5 .9% ,两组疗效差异有非常显著性 (χ2 =9.5 ,P<0 .0 1)。 MTT实验结果与临床化疗结果的符合率 :81.4 8%。结论 改良 MTT法癌细胞体外药物敏感性试验可以预见胃、肠癌的化疗。 Objective To study modified MTT of cancer cell drug sensitivity test and the application of chemotherapy for stomach and intestine cancer under it's direction.Methods The chemotherapy of the modified MTT test group was according to the result of the MTT test in which there were 27 cases of fresh cancer cells samples being density gradienty centrifugation,while the resistence cases in the test group and the experienced control group with gastrointestine cancer were treated based on clinical experienced chemotherapy.Results Among all the 27 cases who finished the MTT test,the results in 8 cases of stomach cancer and 5 cases of intestine cancer were positive,while 7 cases of gastric cancer and 5 cases of intestinal cancer were negative.The percentage of effective chemiotherapy (76.9%) with positive result in modified MTT test group was significantly higher than that of 14.3% with negative results(χ 2=10.7, P <0.01).The total effective rate(44.4%) in test group was significantly higher than that of 25.9% in control group(χ 2=9.5, P <0.01).The coincident percentage between the modified MTT group and the results of the clinical chemotherapy was 81.4%.Conclusion Modified MTT method of cancer cells drug's sensitivity test may anticipate the result of the stomach and intestine cancer chemotherapy.
