摘要
目的:探讨Ⅱ组mGluRs激动剂DCG-Ⅳ对弥漫性脑损伤(DiffuseBrainInjury,DBI)及其合并二次脑损伤(SecondaryBrainInsult,SBI)后损伤脑组织的神经保护作用。方法:单纯DBI及合并SBI后,脑室注射DCG-Ⅳ(100μmol/L,10μl)或生理盐水(10μl),观察大鼠行为、脑组织含水量及皮层损伤神经元数变化。结果:DCG-Ⅳ对DBI后大鼠死亡率(生理盐水组死亡10只,DCG-Ⅳ组死亡9只;P>0.05)、皮层损伤神经元数量及脑组织含水量无明显影响(P>0.05);但可降低合并SBI组大鼠死亡率(生理盐水组死亡20只,DCG-Ⅳ组死亡11只;P<0.05)、减轻合并SBI后的脑水肿程度,减少神经元损伤数量(P<0.05)。结论:DCG-Ⅳ对DBI合并SBI后损伤脑组织有一定的保护作用。
AIM:To study the neuroprotective effects of DCG Ⅳon DBI and SBI. METHODS:All treatment group animals were injected intracerebroventricularly with DCG Ⅳ(100 mmol/L, 10 μl) 1 hour after injuries, while all control group animals were injected with normal saline instead. Then the rats were tested for the motor and cognitive performance 12 hours after injuries. The water contents and the numbers of injured neurons were counted at different intervals post injuries. RESULTS:Although the mortality, the water contents in the frontal cortex, the damaged neurons in the parietal cortex of the rats with DBI alone could not be reduced by the treatment of DCG Ⅳ(the death number of NS group was 10 and DCG Ⅳgroup was 9,P >0.05;), these could be reduced in the rats with DBI coupled with SBI after the treatment(the death number of NS group was 20 and DCG Ⅳgroup was 11,P< 0.05). CONCLUSION:The results of this study indicate that DCG Ⅳprovides remarkable protection against the damage exacerbated by secondary injury.
