摘要
目的 建立血清脂蛋白 (a) [Lp(a) ]单克隆抗体免疫比浊测定法。方法 应用 2 0株载脂蛋白 (a) [Lp(a) ]单克隆抗体 ,于自动分析仪免疫终点法检测Lp(a)。结果 检测试剂与其它Apo和纤维蛋白原及人血清白蛋白均无交叉反应。胆红素 (15 μmol L)和血红蛋白 (2 .5g L)对测定无干扰 ,检测范围 0～ 12 0 0mg L。该法与ELISA对照 ,差异无显著性 ,r =0 .98。批内CV分别为 3 .2 % [Lp(a) 87mg L] ,2 .9% [Lp(a) 2 5 6mg L] ,2 .7% [Lp(a) 65 8mg L] ;批间CV分别为 4.1%、3 .7%和 3 .5 %。结论 建立的免疫比浊测定血清Lp(a)方法 ,具有特异、灵敏、简便。
Objective To establish a immunoturbidimetric assay (ITA) of lipoprotein (a) with monoclonal antibodies.Methods 20 Lp(a) monoclonal antibodies were used and the method suited for automatic analysis using ITA.Results The other Apo and plasminogen and serum albumin of human had not cross reaction with Lp(a). No interference of nonspecific reaction was observed for bilirubin (15 μmol/L) and hemoglobin (2.5 g/L).The measurement range was 0～1 200 mg/L.There was no difference between ITA and ELISA,r=0.98.Inter assay precision was estimated by assaying 20 replicated samples of three pooled sera with low (87 mg/L), middle (256 mg/L) and high (658 mg/L) Lp(a) concentration respectively,and the CV was 3.2%? 2.9% and 2.7%.Inter assay precision was evaluated with the same samples on 20 consecutive work days and resulted in CV of 4.1%,3.7% and 3.5% respectively.Conclusion The immunoturbidimetric assay of serum lipoprotein (a) with monoclonal antibodies possesses such advantages as specificity,sensitivity,rapidity,simplicity and application for automatic analysis.
