摘要 目的 :研究转染抗凋亡基因bcl 2对心肌细胞活力的影响 .方法 :提取含有人bcl 2基因的逆转录病毒真核表达载体 pDOR -SB质粒 ,用脂质体介导的基因转染法分别将pDOR -SB质粒和pDOR质粒 (不含bcl 2基因 )转染心肌细胞 ,观察转染心肌细胞在缺氧环境下的存活能力 .结果 :①原位杂交可见bcl 2基因转染了心肌细胞 ; ②转染bcl 2基因能明显提高心肌细胞在缺氧条件下的存活能力 ,提高对缺氧的耐受力 .结论 :转染bcl AIM: To observe of effects of transfecting a ntiapoptosis gene into cardiomycytes on cell viability in vitro. METHODS: The retrovirus eukaryotic expression vector pDOR-SB c ontaining human bcl-2 gene was extracted and identified by agarose gel electrophoresis after being cut with restricted e ndonuclease EcoRⅠ. The pDOR-SB and empty vector pDOR we re introduced into cardiomyocytes by liposome-mediated gene transfection method. The transfection was identified with in situ hybridizat ion. The growth properties and tolerance to hypoxia of the cardiomyocytes were s tudied in bcl-2 gene transfected cardiac and non-transfected cardiac cells. RESULTS: The in sit u hybridization detection showed that bcl-2 gene was transfected into most of the pDOR-SB transfected cells, whi ch increased the cell viability and hypoxic tolerance of cardiomyocytes. CONCLUSION: Transfecting bcl-2 gene into cardiacmyocytes can significantly enhance the survival of cardiomyocyt es under hypoxic condition.
