摘要
目的 :在TA2小鼠自发乳腺癌肺转移BCML模型上 ,采用了基因芯片技术 ,比较TA2小鼠低转移MA737模型与高转移BCML模型基因表达的差异 ,以期从基因水平上揭示乳腺癌的转移机制并为进一步寻找其新的防治手段打下基础。方法:采用上海联合基因公司提供的含2048个小鼠cDNA的C57BL -6的基因芯片。研究对象为TA2小鼠自发乳腺癌肺转移BCML模型 ,同时设TA2小鼠自发乳腺癌MA737模型为实验对照组。结果 :筛选出包括与DNA结合、转录和转录因子、蛋白翻译、细胞周期、转移等相关的表达差异的基因共457个 ,其中210个表达上调 ,247个表达下调。结论 :乳腺癌在转移过程中存在许多基因表达调控的改变 ,对于相关基因的研究有助于认识乳腺癌转移的分子机制 。
Objective:To investigate the genes differential expressed in tumor cell stem of low metastasis MA737 and high metastasis BCML of TA2 mice spontaneous breast cancer using cDNA microarray.Methods:A series of expression microarray analysis in lung metastasis mould BCML and contrast group mould MA737 were initated by C57BL-six of cDNA microarray which representing a set of 2 048 mice genes.Results: By applying this cDNA microarray 457 differentially expressed genes were identified and were further identified 210 upregulated and 247downregulated genes for functional analysis.Conclusion: DNA microarray technology is an effective technique in screening for differentially expressed genes in the lung metastasis mould BCML of TA2 mice spontaneous breast cancer. Further analysis of the obtained genes will help to understand the molecular mechanism of breast cancer metastasis.
