摘要
目的 :探讨银杏叶提取物 (Gb E)对糖尿病 SD大鼠的肾脏保护作用。方法 :将雄性 SD大鼠建成链脲佐菌素诱导的糖尿病模型。设正常组 (N组 ,6只 )、糖尿病非治疗组 (DM组 ,8只 )、糖尿病用银杏叶提取物治疗组 (DG组 ,8只 )。6周后观察各组大鼠血糖、体重、2 4 h尿蛋白排泄、肾重等生化指标 ,并用免疫组织化学方法观察肾脏 TGF-β1 、C- fos的表达及肾脏的病理形态学改变。结果 :与 DM组相比 ,DG组肾脏肥大指数及 2 4 h尿蛋白排泄量 (P<0 .0 1,<0 .0 5 )、肾组织 TGF- β1 表达 (P<0 .0 1)及 C- fos表达 (P<0 .0 5 )均明显减少 ,同时糖尿病大鼠肾脏病理改变亦减轻。结论 :Gb E对糖尿病肾病有保护作用。
Objective:To investigate the effects of ginkgo biloba extract (GbE) on the protection of renal injury in diabetic SD rat.Methods:Mail SD rats were included in the study. They were divided into three groups at random. N group: normal control group, n =6; DM group: untreated diabetic group, n =8; DG group: GbE treated diabetic group, n =8. Diabetic rats were induced by intraperitoneal streptozotoeia injection (65mg/kg) at one dose.Rats were killed after 6 weeks treatment. Body weight, kidney weight, blood glucose and urinary protein excretion were detected. Furthermore, the protein expression of renal transforming growth factor β 1 (TGF β 1) and immediate early genes (C fos) were determined by immunohistochemistry, and to observe the pathologic changes in kidney.Results:In diabetic rats treated with GbE, the excretion of 24 hour urinary protein, kidney/body weight ratio and the expression of renal TGF β 1 and C fos were decreased significantly as compared to DM group ( P <0.05, P <0.01, P <0.01, P <0.05,respectively). The pathologic changes were light in kidney. Conclusion:GbE provide a new therapeutic approach to attenuate diabetic nephropathy.
