摘要
目的 探讨可溶性CD40L(sCD40L)在再生障碍性贫血 (再障 )患者体内的变化及其对正常人单核细胞凋亡的影响。方法 用ELISA方法检测 40例再障患者及正常人血清中sCD40L水平 ;用流式细胞仪检测以再障患者血清培养后的正常人单核细胞的凋亡率。结果 再障患者血清中sCD40L水平较正常人显著降低 (P <0 .0 0 1) ;以再障患者血清培养后的正常人单核细胞的凋亡率较对照组明显增加。结论 sCD40L降低可能是AA发病过程中具有特征性的改变 ,并可能因此导致单核细胞凋亡显著增加 。
Objective To investigate the alteration of soluble CD40L (sCD40L) level in the patients with aplastic anemia(AA) and its impact on the apoptosis of normal monocytes. Methods Serum level of CD40L in 40 AA patients was detected by enzyme-linked immunosorbent assay(ELISA).Apoptosis of normal monocytes was analyzed by immunofluorescence and flow cytometry. Results The level of sCD40L in the patients′ serum decreases significantly(P<0.001). The apoptosis rate of the monocytes cultured with serum of SAA increased significantly.Conclusion The decreased level of sCD40L may be a characterized alteration of AA; this alteration may also be one of the reasons of the increased apoptosis of normal monocytes, which may make the patients easier to get infection.
