摘要
目的 :观察氯丙嗪 (CPZ)对谷氨酸 (Glu)诱导的大鼠大脑皮质神经细胞兴奋毒性的影响。方法 :用倒置显微镜观察神经细胞形态的变化 ,用Trypanblue染色法观察细胞生存率 ,测定乳酸脱氢酶 (LDH)以定量反映细胞损伤程度。结果 :5 0 μmol·L 1 Glu使神经细胞死亡率升高 ,LDH释放增加 35 1% ;1,10 μmol·L 1 CPZ能改善神经细胞生长 ,降低细胞死亡率 ,减少LDH的释放。结论
Objective:To study the effect of chlorpromazine (CPZ) on glutamate-induced neurotoxicity in rat primary cultures of cerebral cortical neurons. Methods:The effects of drugs glutamate (Glu) and CPZ on neuron morphology were surveyed with the inverted microscope. Trypan blue exclusion method was used to assess the neuronal viability. Assay of lactate dehydrogenase(LDH) in the supernatant served to represent the extent of neuron injury. Results:50 μmol·L -1 Glu enhanced the death of neurons with a corresponding 351% increase in LDH release. 1, 10 μmol·L -1 CPZ was found to reduce neuron death rate and LDH release induced by Glu and the condition of neuron growth was improved. Conclusion:CPZ was shown to have an obvious protective effect on rat cerebral cortical neurons exposed to the neurotoxic agent Glu.
