摘要
近年来,随着细胞化学、细胞免疫学的发展,作免疫酶标、免疫胶体金标的超薄切片越来越多,这种切片要裱在无支持膜铜网上酶标或金标的反应效果才好;另外普通电镜切片裱在无支持膜铜网上,也有图象清晰、分辨率高容易着色、不易污染的优点。这种无支持膜切片(以下简称无膜切片)虽然好,但制作不易,要制作出好的无膜切片,根据我室近三年来制作的2000余例片子的经验来看,要注意以下几点: 一、切片本身的制作过程:无膜切片由于没有膜的支持,容易断裂和皱折。切片太脆就易断,
The advantages of ultrathin section without supporting membrane on grid lie in these respects:(1)may be gained a more clear picture and a higher resolving power under EM; (2)may be coloured easily and reduce some contamination; (3)can be obtained an excellent result in reaction of immunoenzymic and immuno-colloidal-gold staining. We should pay attention to the quality of ultrathinsection, and of grid used, to the skill of scooping and the time of dyeing.
