肺间质纤维化大鼠肺泡巨噬细胞STAT_1的活化及其依赖性免疫应答基因ICAM-1的表达 被引量:15

STAT_1 activation and STAT_1 -dependent immune-response gene ICAM-1 expression in alveolar macrophages of rats suffered from interstitial pulmonary fibrosis

摘要 目的:探讨肺泡巨噬细胞(AM)中STAT1(signal transducersand activators of transcription 1)的活化在大鼠肺间质纤维化中的作用。方法:50只健康雌性Wistar大鼠随机分成博莱霉素(BLM)组和生理盐水(NS)组各25只,气管内分别灌注BLM和NS。用Western-blot法测定AM中STAT1活化的动态变化;用免疫组化染色法测定AM的ICAM-1(intercellular adhesionmolecule-1)的表达。结果:NS组中的AM内只有少许STAT1活化,气管内灌注BLM后1d,AM中STAT1的活化即开始显著增高,于第7天达高峰,以后逐渐下降,但28 d后仍高于NS组(P<0.05)。NS与BLM组的AM中表达ICAM-1的细胞数增高的模式,与AM中STAT1活化的模式相同。AM中STAT1活化的程度与AM中ICAM-1的表达呈正相关(r=0.913,P<0.01),而后者又与肺组织炎症的积分呈正相关(r=0.947,P<0.01)。结论:实验性肺间质纤维化大鼠AM中存在STAT1的异常活化,并参与了肺泡炎和肺纤维化的形成。 AIM: To investigate the role of signal transducer and activator of transcription 1 ( STAT1 ) in alveolar macrophages (AMs) from rats with bleomycin-induced pulmonary fibro-sis. METHODS: Fifty adult female Wistar rats were randomly divided into two groups: One group was intratracheal-ly instilled with bleomycin ( BLM), while another group with 9 g/L NaCI solution (NS). The kinetic change of STAT, activation and intercellular adhesion molecule-1 (ICAM-1) expression in AMs was examined by Western blot and immuno-histochemical staining, respectively. RESULTS: ① STAT, was rarely activated in AMs of NS group. After bleomycin treatment, the STAT, activation of AMs was significantly increased on day 1, climaxed on day 7, and then gradually decreased, but remaining significantly higher than that of NS group on day 28 (P <0.05). ②There was only a few cells ICAM-1 higher than that positively stained in AMs of NS group. After intratracheal instillation of bleomycin, the number of ICAM-1 positive stained cells was significantly increased on day 1, reached its peak values on day 7, and then gradually decreased, and still higher than that of NS group on day 28 (P<0.05). ③There was a significant cor-relationship between STAT, activation and ICAM-1 expression in AMs (r = 0.913, P<0.01). Furthermore, the ICAM-1 expression in AMs was significantly correlated with the severity of inflammation in lung tissues (r = 0.947, P<0.01). CONCLUSION; STAT, was abnormally activated in AMs of rats with bleomycin-induced interstitial pulmonary fibrosis (IPF). The abnormal STAT, activation may play a role in the pathogenesis of acute alveolitis and pulmonary fibrosis.
