摘要
为探讨肺动脉高压时内源性血管弹性蛋白酶 (EVE)的动态变化及其与肺血管重建之间的关系 ,利用野百合碱建立大鼠肺高压模型 ,于实验第2、8、14、21天 ,使用荧光分光光度计检测肺动脉的弹性蛋白离解活性 ,通过右心导管法测定肺动脉压力 ;并在上述时间点 ,对周围肺动脉进行组织病理学分析。结果显示 ,肺动脉的弹性蛋白离解活性在注射野百合碱后的第2天即明显升高(3.87±0.45) ,第8天降为正常 (0.18±0.02) ,第14天再次出现升高现象 (1.45±0.18) ,并一直持续到第21天(1.91±0.22) ;而肌性肺动脉的比例增加在注射野百合碱后第8天出现(32.67±4.23) ,肺动脉中膜的增厚 (4.20±0.37)及肺动脉压力的增高 (21.00±2.10)在第14天后出现。提示EVE活性的早期升高可能是肺动脉高压的一种重要触发因素 ;
to explore the relationship between the dynamic changes of elastoLyTic activity and pulmonary remodeling in rats with pulmonary hypertension induceD by monocrotaline,the rat models with pulmonary hypertension were induced by sUbcutaneous injection of monocrotaline.the elastolytic activity was measured witH a fluorescence microplate reader and pulmonary artery pressure detected with cArdiac catheterization on days2,8,14and21.the light microscopic structural anaLysis of the peripheral pulmonary vasculature was performed at the same time poiNts.the results showed that elastolytic activity increased significantly on day2,then returned normal on day8,but elevated again on day14,and kept on until dAy21.extension of muscle into distal arteries was observed on day8.increased medIal wall thickness and pulmonary artery pressure were present on day14.all this PHenomena indicate that the early increase of elastolytic activity may be related to the initiation of the pulmonary hypertension and the next increase may be inVolved in the development of this disease.
