摘要
目的 观察单胺递质在抗抑郁剂产生的效应中所起的作用。方法 单胺递质 (5 HT、DA、NE)、去甲丙咪嗪(DIM)及MK80 1与皮质酮 0 2mmol·L-1共孵PC12细胞48h ,并用MTT比色法判断细胞损伤程度。采用Fura 2 /AM荧光标记法测定胞内Ca2 + 浓度。结果 高浓度皮质酮处理PC12细胞后 ,细胞存活率比正常组降低 ,胞内 [Ca2 + ]i升高 ,表明细胞受到损伤或部分死亡。当给予 5 HT ,DA ,NE ,DIM及MK80 1后 ,细胞存活率增高 ,而胞内 [Ca2 + ]i降低 ,细胞损伤减少或已接近正常。结论 与抗抑郁剂一样 ,单胺递质对皮质酮损伤的PC12细胞有明显的保护作用 ,并且能够明显降低皮质酮诱发的PC12细胞胞内Ca2 + 超载。提示单胺递质可能参与或介导了抗抑郁剂的神经元保护作用 。
AIM To evaluate the mechanism of antidepressant action. METHODS Cultured PC12 cells were treated with corticosterone(Cort) 0 2 mmol·L -1 , at the presence of monoamines, desipramine (DIM), or MK801. The cellular viability and intracellular free Ca 2+ concentration ([Ca 2+ ] i)were determined by using MTT assay and Fura 2/AM fluorescence labelling respectively. RESULTS After incubation with Cort 0 2 mmol·L -1 for 48 h, the cellular viability was decreased and the [Ca 2+ ] i was overloaded, suggesting that Cort induced lesion in the PC12 cells. While at the presence of 5 HT, DA, NE, DIM, or MK801, the cellular viability was all improved and the [Ca 2+ ] i overload was attenuated, indicating that treatment with monoamines or antidepressant DIM protected PC12 cells lesioned by Cort. CONCLUSION Just the same as DIM, monoamines all antagonized the lesion induced by Cort in PC12 cells, which maybe related to that they attenuated the [Ca 2+ ] i overload induced by Cort. These results extended the evidence that monoamines participate and mediate the antidepressant action in details.
