摘要
目的初步探讨建立人胚腺垂体细胞库的方法及冷冻移植物复温后的形态和功能。方法 将人胚腺垂体细胞培养至第5天冷冻,15天后复温再培养,与单纯培养组相对照,相差显微镜下观察细胞形态,苔盼蓝测拒染率及检测激素分泌功能。结果 冷冻复温后垂体细胞存活率平均达78.2%;光镜下生物学状态无改变;GH分泌细胞复温后第5-7天状态最佳;PRL、TSH分泌水平差。结论本方法适于建立人胚腺垂体细胞库用于治疗垂体性侏儒症,移植时机为复温后5-7天。该冷冻方法不适用于PRL分泌细胞,培养条件不适用于TSH分泌细胞。
Objective To study the method of foundation of human fetal adenohypophysial cell bank and the morphology and function of frozen-thawed(F-T)graft. Methods The human fetal adenohypophysial cells that were freezed when culturing for 5 days were thawed and recultured after 15 days. Compared to the simple culture group, they were monitored in the microscope and assayed by Typan blue and RIA. Results About 78. 2% F-T cells were survived and similar to primary cell in growing condition in the microscope. The GH secretion was the highest and the cell growing was best in the 5 ~ 7th day while the PRL and TSH secretion was low.Conclusion The way of this experiment is suitable to build the human fetal adenohypophysial cell bank for treating pituitary dwarfism. We should carry out clinic transplantation when the F-T cell have recultured for 5-7 days.The method is unfit for PRL secretion in freezing condition and for TSH secretion in culturing condition.
