摘要
目的 探讨扩张型心肌病 ( DCM)易感的分子机制和确立一种较为实用的 MHC- 类基因的检测方法。方法 采用聚合酶链式反应和顺序特异性引物 ( PCR- SSP)基因分析方法 ,对 31例扩张型心肌病患者及 30例无血缘关系的健康人的人类主要组织相容性复合体 ( MHC- 类 ) DQ各等位基因及亚基因进行了检测分析 ,并将该方法与其它检测 MHC- 类基因的方法进行对比。结果 结果表明 ,MHC- DQB1* 0 30 1基因与 DCM呈正相关 ( P <0 .0 1) ,其它 MHC- DQB1*其它各等位基因未见异常。结论 MHC- DQB1* 0 30 1基因可能为北方汉族人 DCM的致病易感基因。尽管目前 MHC- 类基因分型方法已有多种 ,但所采用的方法 ( PCR- SSP)具有快速、简便、敏感、准确和可靠等优点。
Objective In order to study the correlativity between MHC-DQB1 gene polymorphism and Dilated Cardiomyopathy (DCM).Methods MHC-DQB1 gene polymorphism in 31 patients with DCM and 30 normal control subjects was analyzed by using the polymerease chain reaction sequence specific primer (PCR SSP)technique.Results The frequency of MHC-DQB1 0301 allele was significantly higher in patients than that in the controls,RR of DCM patients was,P<0.01.Conclusions These findings further verify that immunogenetics play a predominant pathogenetics role in a subset of DCM patients.Individuals carrying MHC-DQB1 0301 alleles may be susceptibility gene to DCM. This method is More specific, sensitive, simple,rapid and accurate,and could be applied to the routine clinical detective.
