三氧化二砷对人结肠癌细胞增殖及凋亡的影响 被引量:6

摘要 目的 观察三氧化二砷 (As2 O3 )对人结肠腺癌LS 174T细胞增殖及凋亡的影响。方法 应用MTT法、细胞集落形成试验、流式细胞仪技术和TUNEL法观察不同浓度的As2 O3 (1.0、2 .0、4 .0、8.0、16 .0 μmol/L)对LS 174T细胞增殖和凋亡的影响 ,免疫组化技术观察As2 O3 对bcl 2基因表达的影响。结果 As2 O3 处理后 ,LS 174T细胞生长抑制率上升 ,集落形成率下降 ,细胞周期中G1期细胞比例上升、S期和G2 /M期细胞比例下降 ,TUNEL法显示凋亡细胞数增加 ,免疫组化结果显示bcl 2的表达明显减弱。结论 As2 O3 能抑制LS 174T细胞增殖并诱导该细胞株凋亡 ,其机理可能与下调bcl Objective To study the effect of As 2O 3 on apoptosis and bcl 2 expression in LS 174T human colon cancer cells. Methods The inhibitive effect of As 2O 3 on LS 174T was measured with MTT. Colony forming rate of LS 174T treated with As 2O 3 were studied by colony forming assay.Apoptosis of LS 174T was studied by TUNEL.The expression of bcl 2 gene was determined by immunohistochemical analysis. Result After treatment by As 2O 3,the cell line's inhibition index increased while the colony forming rate decreased,the number of the apoptosis cell increased,the prorortion of G 1 phase increased while the G 2/M and S phase decreased in the cell cycle. In As 2O 3 treated cell line,bcl 2 expression was down regulated at protein level. Conclusion As 2O 3 inhibited the proliferation of LS 174T by inducing apoptosis in the study. Down regulation of bcl 2 protein may be the mechanism.
