摘要
目的 探讨当归多糖 (AP- 0 )及其硫酸酯 (APS)对凝血和血小板聚集的影响。方法 红外比浊法测定血小板聚集率、凝血酶原时间 (PT)、凝血酶时间 (TT)、活化部分凝血活酶时间 (APTT) ;断尾法测出血时间 ;玻片法测凝血时间 ;肝素抗凝测血液流变学指标。结果 当归多糖及其硫酸酯显著延长凝血时间 ,缩短出血时间 ;显著性延长 TT和 APTT,而对 PT影响较小 ;显著升高 5 min血小板聚集率 ;AP- 0显著升高低切全血粘度 (ηb) ,增强红细胞 (RBC)的聚集性 (AI) ;而 APS显著降低高、低切变率 ηb、血浆粘度 (PV)和 AI。结论 当归多糖及其硫酸酯有较强的抗凝血和止血作用 ,其止血作用与其促进血小板聚集作用有关 ;
Object To investigate the effects of Angelica polysaccharide (AP 0) and its sulfates (APS) on blood coagulation and platelet aggregation. Methods Infrared turbidimetric method was used to estimate platelet aggregation, prothrombin time (PT) and thrombin time (TT), actiated partial thromboplastin time (APTT); bleeding time (BT) was measured by cutting the mouse's tail and coagulate time (CT) was measured by glass method sheet; hemorheology was measured with heparin as anticoagulant.Results The AP 0 and APS prominently enhanced the platelet aggregation at 5 min (PAG ), while showed less effects on the maximum platelet aggregation (PAG ); they, especially APS 1, also markedly prolonged CT but shortened BT, both AP 0 and APS 1 significantly prolonged TT and APTT in all experimental dosage, while only APS 1 in high dosage (8.46 mg/kg) significantly prolonged PT; AP 0 increased the aggregation index (AI) of RBC and the whole blood viscosity (η b) at shear rate of 5 s -1 at a dose of 2.00 mg/kg, however, unlike AP 0, APS 1 decreased AI and η b at shear rate of 5 s -1 and 230 s -1 . Conclusion These results suggest that AP 0 and APSs have potent anticoagulant and haemostasis effects. The haemostasis effect is related to promoted platelet aggregation. Ameliorative effect of Angelica polysaccharide sulfate on hemorheology is better than that of Angelica polysaccharide itself.
