鸡肾脏胰岛素样生长因子-Ⅰ分解酶特性研究

摘要 以12 5Ⅰ标记的、人工合成的人胰岛素样生长因子Ⅰ (IGF Ⅰ )为底物 ,以鸡肾脏的提取液为粗酶液 ,研究在不同时间、粗酶液浓度、温度及pH值等条件下的酶促分解反应情况。结果显示 :①IGF Ⅰ分解酶反应时间以 30min为宜 ;②粗酶液的含量宜在 1 0 8μg·μL-1以下 ;③酶活性在 4 5℃时最高 ;④此酶在 5 0℃以下稳定 ,当温度升至 5 5℃时其活性急剧下降 ,升至 6 0℃时将降至最大活性的 2 0 %左右 ;⑤此酶最适pH值为 6 5~ 7 0。 Chicken insulin like growth factor Ⅰ(IGF?Ⅰ)labeled by 125 I was incubated with IGF?Ⅰ degradation enzyme(crude extraction from kidney)under various conditions,such as incubation time,temperature,pH,and content of crude enzyme.When the reaction stopped,the mixture was centrifuged,the CPM(counts per minute)of supernatant indicated the activity of IGF?Ⅰ degradation enzyme.The results showed:1)The optimum incubation time of the enzyme was 30 min.2)The suitable content of crude enzyme was below 1 08 μg·μL -1 .3)The enzyme had maximum activity at 45 ℃.4)Below 50 ℃ the activity of enzyme had no change;but if the temperature reached 55 ℃,it dropped suddeny;at 60 ℃ it dropped to about 20% of the highest activity.5)The most appropriate pH for the enzyme was 6 5?7 0.
