摘要
从乔川黄酒麦曲中分离产蛋白酶霉菌,对分离到的霉菌进行形态学观察、生理生化试验,对高产蛋白酶菌株进行了筛选、分子生物学鉴定及生长特性试验。共分离到7株霉菌,分别命名为M1,M2,M6,M7,M9,M12,M13,形态学鉴定为毛霉属。7株菌均产蛋白酶,产蛋白酶活力最高的菌株为M2和M7,分子生物学鉴定其分别为总状横梗霉属(Lichtheimia ramosa)、卷枝毛霉(Mucor circinelloides)。生长特性研究结果表明,M2和M7生长最适pH值为5.0,其他环境下其生长速度均会受到抑制;酒精耐受力研究结果表明,乙醇体积分数越高,M2和M7生长速度越低,乙醇体积分数大于3%时,其生长会受到显著抑制。
Seven protease-producing molds were isolated from Maiqu(traditional fermentation starter)of Qiaochuan yellow rice wine which named M1,M2,M6,M7,M9,M12 and M13.Morphological observation and biochemical characterization revealed that all isolated strains exhibited protease activity,with strains M2 and M7 showing the highest enzymatic activities.Molecular identification demonstrated strain M2 as Lichtheimia ramosa and strain M7 as Mucor circinelloides.Growth optimization studies indicated an optimal pH of 5.0 for both strains,with significant growth inhibition observed at pH values deviating from this optimum.Ethanol tolerance tests showed that growth rates decreased proportionally with increasing volume concentration of alcohol,reaching complete inhibition at concentrations above 3%.
