摘要
本研究建立一种快速检测香草兰豆荚中香兰素的超临界流体色谱法:采用ACQUITY UPC2TM BEH 1.7μm(3.0×100 mm Column)色谱柱,流动相为二氧化碳和甲醇(二氧化碳:甲醇=95.0:5.0),背景压力为2000 psi,流速为1.5 mL/min,进样量1.0μL,柱温箱为55℃,检测波长275 nm。结果表明,香兰素最快可在0.678 min出峰,本方法在21.1~337.6μg/mL浓度时线性关系良好,相关系数R^(2)为0.9995,仪器检测限为1.79μg/mL,定量限为5.97μg/mL,加标回收率为90.50%~95.55%。与现行液相仪器检测方法相比,本方法的检测效率可提高10倍,进样量可降低10倍,有机试剂等流动相可降低20倍,运行成本降低50倍以上,在省时、高通量、低毒和生态友好方面具有独特的优势,可用于香草兰中香兰素的检测。
In this study,a rapid method for the determination of vanillin in vanilla pods was established using supercritical fluid chromatography.Chromatographic separation was performed on an ACQUITY UPC2TM BEH column(1.7μm,3.0×100 mm)with carbon dioxide and methanol as the mobile phase(CO_(2):methanol=95:5).The back pressure was maintained at 2000 psi,the flow rate was 1.5 mL/min,the injection volume was 1.0μL,the column temperature was 55°C,and the detection wavelength was 275 nm.The results showed that vanillin could be detected at a retention time as short as 0.678 min.A good linear relationship was obtained within the concentration range of 21.1~337.6μg/mL with a correlation coefficient R^(2) was 0.9995.The limit of detection(LOD)and limit of quantification(LOQ)were 1.79μg/mL and 5.97μg/mL,respectively.The recovery rate ranged from 90.50%to 95.55%.Compared with the current liquid chromatography methods,this method improved detection efficiency by 10 times,reduced the injection volume by 10 times,decreased the organic solvents in the mobile phase by 20 times,and lowered operating costs by more than 50 times.This method had unique advantages in terms of time-saving,high throughput,low toxicity and environmental friendliness,and can be applied to the determination of vanillin in vanilla pods.
