摘要
研究旨在通过网络药理学和分子对接技术系统地探究植物乳杆菌(Lactiplantibacillus plantarum,Lp)治疗仔猪腹泻的活性成分及其潜在作用机制。基于Lipinski五规则筛选Lp代谢产物中的活性成分,结合Swiss Target Prediction数据库预测潜在靶点,并通过GenCards和OMIM数据库获取仔猪腹泻相关靶点。利用Venny 2.0.2分析获得交集靶点;通过STRING数据库构建蛋白质互作(PPI)网络,进行基因本体论(GO)功能注释和京都基因组百科全书(KEGG)通路富集分析(P<0.05);最后通过AutoDock Tools进行分子对接验证。研究共筛选出46个活性成分(包括5-羟基吲哚乙酸、异吡啶磺胺、酪氨酸等)和356个交集靶点,核心靶点涉及EGFR、ALB、MMP9等。GO和KEGG分析显示,Lp可能通过神经活性配体-受体相互作用、钙信号通路等生物过程缓解腹泻。分子对接结果显示,异吡啶磺胺与MMP9具有较强的结合亲和力。结果表明,植物乳杆菌可能通过多成分-多靶点-多通路机制缓解仔猪腹泻,为深入理解其益生机制及进一步开发应用提供了理论依据。
This study aimed to systematically explore the active components of Lactiplantibacillus plantarum(Lp)in treating piglet diarrhea using network pharmacology and molecular docking technology.The active components in the metabolites of Lp were screened based on the Lipinski’s Five Rules,and the potential targets were predicted by the Swiss Target Prediction database.Meanwhile,the piglet diarrhea-related targets were obtained from the GenCards and OMIM databases.The intersection targets were analyzed by Venny 2.0.2.The protein-protein interaction(PPI)network was constructed using the STRING database,followed by Gene Ontology(GO)functional annotation and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis(P<0.05).Finally,molecular docking verification was performed via AutoDock Tools.A total of 46 active components(including 5-hydroxyindoleacetic acid,isopyridinylsulfonamide,tyrosine,etc.)and 356 intersection targets were screened out,with the core targets including EGFR,ALB,MMP9 and so on.GO and KEGG analyses showed that Lp might alleviate diarrhea through biological processes such as neuroactive ligand-receptor interaction and calcium signaling pathway.Molecular docking results indicated that isopyridinylsulfonamide had strong binding affinity with MMP9.It is concluded that Lactiplantibacillus plantarum may alleviate piglet diarrhea through a multi-component,multi-target and multi-pathway mechanism,which provides a theoretical basis for further understanding its probiotic mechanism and further development and application.
