摘要
目的 探讨秦皮乙素(ESC)对T细胞增殖的抑制作用及相关机制。方法 分离纯化人T细胞,利用抗白细胞分化抗原(CD)3/CD28抗体或植物血凝素(PHA)将其激活,将细胞分为空白组、T细胞活化组、磷脂酰肌醇3-激酶(PI3K)抑制剂2-(4-吗啉基)-8-苯基-4H-1-苯并吡喃-4-酮(LY294002)药物对照组及ESC(10、20、40μmol/L)处理组。利用流式细胞术检测细胞的增殖、凋亡、存活率、CD25表达水平及细胞周期;采用酶联免疫吸附试验(ELISA)检测细胞分泌的白细胞介素(IL)-2、IL-6、IL-17A水平。结果 与T细胞活化组相比,ESC处理组的活化T细胞增殖数量明显降低(P<0.05);两组在活化T细胞凋亡率及CD25表达水平方面比较,差异无统计学意义(P>0.05);ESC处理组与空白组的静息T细胞存活率比较,差异无统计学意义(P>0.05)。相较于T细胞活化组,ESC处理组中处于G0/G1期的细胞占比及IL-2水平明显升高,IL-6、IL-17A水平明显下降(P<0.05)。结论 ESC可抑制T细胞增殖,阻滞细胞周期于G0/G1期,并抑制促炎因子IL-6、IL-17A的分泌。
Objective To explore the inhibitory effect of esculetin(ESC)on T-cell proliferation and its related mechanisms.Methods Human T cells were isolated and purified,and then activated using anti-cluster of differentiation(CD)3/CD28 antibodies or phytohemagglutinin(PHA).The cells were divided into a blank control group,a T cell activation group,a phosphoinositide 3-kinase(PI3K)inhibitor 2-(4-morpholinyl)-8-phenyl-4H-1-benzopyran-4-one(LY294002)drug control group,and ESC-treated groups(10,20,40μmol/L).Flow cytometry was used to detect cell proliferation,apoptosis,viability,CD25 expression level,and cell cycle.Enzyme-linked immunosorbent assay(ELISA)was employed to measure the secretion levels of interleukin(IL)-2,IL-6,and IL-17A by the cells.Results Compared with the T cell activation group,the number of proliferating activated T cells in the ESC-treated groups was reduced(P<0.05).There was no statistically significant difference in the apoptosis rate of activated T cells or the expression level of CD25 between the two groups(P>0.05).In addition,there was no statistically significant difference in the survival rate of resting T cells between the ESC-treated groups and the blank control group(P>0.05).Compared with the T cell activation group,the proportion of cells in the G0/G1 phase and the secretion level of IL-2 were upregulated in the ESC-treated groups,while the secretion levels of IL-6 and IL-17A were downregulated(P<0.05).Conclusion Esculetin can inhibit T-cell proliferation,arrest the cell cycle at the G0/G1 phase,and suppress the secretion of pro-inflammatory cytokines IL-6 and IL-17A.
