摘要
为研究结核分枝杆菌蛋白Rv3874(培养滤液蛋白10,culture filtrate protein 10,简称Cfp10)对巨噬细胞鞘脂代谢及相关功能的影响,以非致病性牛型分枝杆菌(BCG)为载体,构建表达Rv3874的重组菌,利用该重组菌感染小鼠巨噬细胞RAW264.7。采用代谢组学及分子生物学等技术,观察Rv3874对巨噬细胞鞘脂代谢通路的影响及相关物质的表达变化,以及这些变化对巨噬细胞免疫活性的潜在影响。结果表明:Rv3874能够改变巨噬细胞鞘脂代谢方向,上调鞘磷脂合酶2(Sgms2)、神经酰胺激酶(Cerk)和半乳糖神经酰胺酶(Galc)等鞘脂代谢关键酶的表达,使其向增加神经酰胺和鞘磷脂的合成方向富集。Rv3874可能通过改变巨噬细胞鞘脂代谢过程进而影响巨噬细胞功能。
To study the effects of Mycobacterium tuberculosis protein Rv3874(culture filtrate protein 10,Cfp10)on sphingolipid metabolism and related functions of macrophages,nonpathogenic Mycobac-terium Bovis(BCG)was used as the vector to construct a recombinant strain expressing Rv3874.The mouse macrophage RAW264.7 was infected with the recombinant strain.Using metabonomics and mo-lecular biology techniques,the effects of Rv3874 on the sphingolipid metabolic pathway of macro-phages and the expression changes of related substances were observed,as well as the potential ef-fects of these changes on the immune activity of macrophages.Rv3874 could change the direction of sphingolipid metabolism of macrophages,upregulate sphingomyelin synthase 2(Sgms2),ceramide ki-nase(Cerk),and galactosylceramidase(Galc)and other key enzymes of sphingolipid metabolism,and enrich them in the direction of increasing the synthesis of ceramide and sphingomyelin.Rv3874 may affect the function of macrophages by changing the reprogramming of sphingolipid metabolism.
