摘要
本研究以巨花鸢尾‘明朝宝藏’花葶、未开花子房、根茎幼芽为外植体通过组培诱导的方式培养再生植株的过程。外植体经预处理消毒后接种于培养基上,结果表明花葶为最佳外植体,成活率为47.5%,当愈伤组织诱导培养激素选择6-BA、IBA时诱导率高达55.67%。而根茎幼芽直接诱导不定芽时效果显著,诱导率达32%。当不定芽长至3~5 cm,转接纯MS培养基中进行生根培养,最早5 d即有小根出现,生根率达100%。将生根良好长势健壮的组培苗移栽至蛭石∶珍珠岩∶营养土为1∶1∶3的混合基质,成活率可达87%。
This study established a plant regeneration system for Iris ampliflora‘Ming Treasure'through tissue culture induction using scapes,unflowered ovaries,and rhizome buds as explants.After pre-treatment and sterilization,the explants were inoculated onto culture media.The results showed that scapes were the most suitable explant,with a survival rate of 47.5%.When 6-BA and IBA were used as phytohormones for callus induction,the induction rate reached 55.67%.Direct induction of adventitious buds from rhizome buds was also effective,achieving an induction rate of 32%.When the adventitious buds grew to 3~5 cm,they were transferred to a hormone-free MS medium for rooting.Root primordia emerged as early as 5 days,with a rooting rate of 100%.Well-rooted and vigorous tissue-cultured plantlets were then transplanted into a mixed substrate with a vermiculite:perlite:nutrient soil ratio of 1:1:3,resulting in an acclimatization survival rate of 87%.
