电针改善老龄小鼠术后认知功能的机制与海马神经元突触可塑性的关系

Relationship between mechanism of electroacupuncture in improving postoperative cognitive function and hippocampal neuronal synaptic plasticity in aged mice

摘要 目的评价电针改善老龄小鼠术后认知功能的机制与海马神经元突触可塑性的关系。方法SPF级健康雄性C57BL/6小鼠96只,16~17月龄,体质量24~26 g,采用随机数字表法分为4组(n=24):对照组(C组)、手术组(S组)、电针非穴位+手术组(NES组)和电针穴位+手术组(ES组)。行剖腹探查术建立小鼠术后认知功能障碍模型;ES组选取百会穴和大椎穴进行电针刺激,NES组于百会穴和大椎穴旁3 mm处进行电针刺激,选用疏密波,频率2/15 Hz,强度1 mA,持续30 min,1次/d,于术前1 d开始连续刺激8 d。分别于术前1 d和术后3、7 d时行Morris水迷宫实验测试认知功能。分别于术后3和7 d水迷宫测试结束后,处死小鼠取海马组织,采用高尔基染色法计数树突分支,计算树突分支长度和树突棘密度,采用蛋白质印迹法测定海马CA1区成熟脑源性神经营养因子(mBDNF)和脑源性神经营养因子前体(proBDNF)的表达。结果与C组比较,S组、NES组和ES组术后3和7 d时逃避潜伏期延长,穿越原平台位置次数减少,海马组织proBDNF表达上调,mBDNF表达下调,mBDNF/proBDNF比值降低,神经元树突长度缩短,树突分支减少,树突棘密度降低(P<0.05);与S组比较,ES组术后3和7 d时逃避潜伏期缩短,穿越原平台位置次数增加,海马组织proBDNF表达下调,mBDNF表达上调,mBDNF/proBDNF比值升高,神经元树突长度和树突分支增加,树突棘密度升高(P<0.05),NES组上述各指标差异无统计学意义(P>0.05)。结论电针改善老龄大鼠术后认知功能的机制可能与增强海马神经元突触可塑性有关。 Objective To evaluate the relationship between the mechanism by which electroacupuncture improved postoperative cognitive function and hippocampal neuronal synaptic plasticity in aged mice.Methods Ninety-six healthy male C57BL/6 mice,aged 16-17 months,weighing 24-26 g,were divided into 4 groups(n=24 each)using a table of random numbers:control group(C group),surgery group(S group),electroacupuncture non-acupoint+surgery group(NES group),and electroacupuncture acupoint+surgery group(ES group).A model of postoperative cognitive dysfunction was established using abdominal surgery in anesthetized mice.Baihui(GV20)and Dazhui(GV14)acupoints were electrically stimulated with sparse-dense waves at the frequency of 2/15 Hz and intensity of 1 mA for 30 min once a day for 8 consecutive days starting from 1 day before operation in group ES,and electroacupuncture was performed at the points 3 mm lateral to the Baihui and Dazhui acupoints instead in group NES.The Morris water maze test was performed to assess cognitive function at 1 day before surgery and 3 and 7 days after surgery.After the end of the Morris water maze test on 3 and 7 days after surgery,the mice were sacrificed,and hippocampal tissues were removed for determination of the count of dendritic branches(using Golgi staining)and expression of mature brain-derived neurotrophic factor(mBDNF)and precursor brain-derived neurotrophic factor(proBDNF)in the hippocampal CA1 region(by Western blot).The dendritic length and dendritic spine density were calculated.Results Compared with group C,the escape latency was significantly prolonged,the number of crossing the original platform was decreased,the expression of proBDNF in hippocampal tissues was up-regulated,the expression of mBDNF was down-regulated,the ratio of mBDNF/proBDNF was decreased,the length of neuronal dendrites was shortened,the number of dendritic branches was decreased,and the density of dendritic spines was decreased at 3 and 7 days after operation in S,NES and ES groups(P<0.05).Compared with group S,the escape latency was significantly shortened,the number of crossing the original platform was increased,the expression of proBDNF in hippocampal tissues was down-regulated,the expression of mBDNF was up-regulated,the ratio of mBDNF/proBDNF was increased,the length of neuronal dendrites and the number of dendritic branches were increased,and the density of dendritic spines was increased at 3 and 7 days after operation in group ES(P<0.05),while no significant changes were found in the aforementioned parameters in group NES(P>0.05).Conclusions The mechanism by which electroacupuncture improves postoperative cognitive function may be related to the enhancement of hippocampal neuronal synaptic plasticity in aged mice.
