摘要
对泸州某酒厂浓香型白酒生产车间连续使用的老窖池窖泥进行富集培养,从中筛选出2株产己酸和丁酸的菌株Ly-1和Ly-2。经形态和分子鉴定,菌株Ly-1和Ly-2均隶属于赖氨酸芽孢杆菌属(Lysinibacillus)微生物,Ly-1为细长赖氨酸芽孢杆菌(Lysinibacillus macroides),Ly-2为解木糖赖氨酸芽孢杆菌(Lysinibacillus xylanilyticus);在最佳培养条件下,两株菌株按照一定比例混合发酵产己酸量(1.28 g/L)比单一发酵(平均0.71 g/L)下的己酸产量高80%;通过单因素试验,选取乙醇添加量、黄水添加量和接种量3个对己酸产量影响较大的因素进行响应面优化,得到最佳条件为乙醇添加量1.9%、黄水添加量3.1%、接种量9.9%;在此条件下,混合菌液的己酸产量达到3.11 g/L。以上复合发酵菌液为企业技术中心进行人工窖泥制备和窖池养护提供了理论支持,将其应用于实际生产,有助于人工窖泥老熟和老化窖泥复壮。
Through continuous enrichment culture of aged pit mud from a Nongxiang Baijiu distillery,two strains that produce hexanoic acid and butyric acid were screened out and named Ly-1 and Ly-2.Morphological and molecular identification indentified Ly-1 as Lysinibacillus macroides and Ly-2 as Lysinibacillus xylanilyticus.Under the optimal cultivation conditions,the mixed fermentation of the two strains yielded 1.28 g/L of caproic acid,80%higher than single-strain fermentation(average 0.71 g/L).Single-factor experiments were conducted,and the amount of ethanol,the amount of pit bottom water,and the inoculation volume were selected as the three main factors influencing the production of caproic acid.The optimal parameters were determined through response surface method as follows:the amount of ethanol was 1.9%,the amount of pit bottom water was 3.1%,and the inoculation volume was 9.9%.Under these conditions,the caproic acid content of the mixed bacterial solution reached 3.11 g/L.This study has provided theoretical support for the preparation of artificial pit mud and pit maintenance.The application of the mixed bacterial solution in actual production can facilitate the maturation of artificial pit mud and rejuvenation of aged pit mud.
